Establishment and multiplication in vitro of clones of Eucalyptus dunnii and Eucalyptus benthamii using light-emitting diodes (LEDs)

Abstract

Tissue culture has been presented as one of the main techniques in vegetative propagation, highlighting the importance of measures in controlling and preventing contamination. The use of light-emitting diodes (LEDs), and certain colors, can be highlighted as an aid for the establishment and multiplication of disinfested explants. The specific objectives of the present chapter had were the disinfestation of explants belonging to different clones of Eucalyptus dunnii and Eucalyptus benthamii submitted to different immersion times and concentrations of sodium hypochlorite (NaOCl), ranging from 0.5 to 1.5% and 10 to 20 minutes respectively. Also the establishment of explants aiming fot their multiplication in vitro, using LEDs of blue, red and mixed coloration. The use of NaOCl at a concentration of 1.5% with immersion for 10 minutes proved to be the most effective method of disinfesting the material. As for the establishment and multiplication of explants, the use of blue light LEDs resulted in a higher rate of sprouting during establishment and a greater appearance of calluses (no sprouting at this stage) during multiplication.
